There are two schools of thought here.
Shortly before cooking: Ideal for quick cooking such as steaks or hamburgers. Apply the rub, let it 'sweat' for 15-20 minutes (you will see the moisture of the meat dissolve the spices) and then go on the grill.
Well in advance (Dry Brining): For large cuts such as brisket or pork shoulder, applying the rub (especially if based on salt such as SPG+) many hours before, or even the night before, allows the salt to penetrate deep into the meat, flavoring it and keeping it more moist during long cooking. This process is called 'dry brining' (dry brine).
